Abstract:
The composition of biological samples is complicated, the protein and other macromolecules should be removed at first before analysis to avoid damaging the chromatographic column. The common methods used for pre-treatment are multi-line operation and time-consuming, furthermore cause damage and contamination of the sample. The method of direct injection can reduce the sample pretreatment step, effectively purify and pre-concentrate the sample. This process can be automated by column-switching technique, it will improve the reproducibility of the method. In this paper, automated processing of current progress of sample in liquid chromatography analysis are reviewed, especially focus on the direct injection technologies such as restricted access material and other automated online solid phase extraction process, also include the more selective emerging automated sample extraction phase techniques, such as molecular imprinted polymers. Finally, these methods and potential applications are briefly discussed.
