Abstract:
OBJECTIVE To establish an analytic method of multi components of Huai Chrysanthemi Flos by HPLC and its related fingerprint pattern recognition. METHODS HPLC was used to measure the content of chlorogenic acid, 3,4-dicaffeoyl puininic acid, 3,5-dicaffeoyl puininic acid, 4,5-dicaffeoyl puininic acid, luteolin and establish the fingerprint of Huai Chrysanthemi Flos. The fingerprint recognition pattern was established by using principle component analysis and clustering analysis based on the results from 10 batches of Huai Chrysanthemums Flos. Subsequent similarity evaluation was conducted by using similarity evaluation software. The column was SWELL Chromplus C
18 (250 mm×4.6 mm, 5 μm). The mobile phase was acetonitrile-0.3% formic acid. Gradient elution was used and its flow rate was 0.8 mL·min
-1. Injection volume was 10 μL. Detection wavelength was 350 nm. Column temperature was 25?℃. RESULTS The average content ranges for chlorogenic acid, 3,4-dicaffeoyl puininic acid, 3,5-dicaffeoyl puininic acid, 4,5-dicaffeoyl puininic acid and luteolin were 0.04%-0.27%, 0.01%-0.21%, 0.45%-1.09%, 0.32%-0.96% and 0.02%-0.18%, respectively. The fingerprint pattern recognition of Huai Chrysanthemi Flos was established, and there were 15 common peaks in the identified fingerprints. The samples were divided into five categories based on the first three principal components extracted in fingerprints. CONCLUSION This method is simple, accurate and reproducible, which can provide a reference for the multi-component determination of Huai Chrysanthemi Flos, thus providing the scientific evidence for its quality control.
