Simultaneous Determination of Three Components in Bai Lemian Capsule by HPLC

摘要

摘要: 目的建立同时测定百乐眠胶囊中2,3,5,4＇-四羟基二苯乙烯-2-O-β-D葡萄糖苷、远志口山酮Ⅲ、3,6二芥子酰基蔗糖的高效液相色谱法。方法采用Agilent Eclipse XDB-C₁₈色谱柱(4.6 mm×250 mm，5 mm)；流动相：甲醇-乙腈-0.05%磷酸溶液(32∶8∶60)；流速：1.0 mL·min^-1；检测波长：320 nm。结果 2,3,5,4＇-四羟基二苯乙烯-2-O-β-D葡萄糖苷、远志口山酮Ⅲ和3,6二芥子酰基蔗糖线性范围分别为0.020 6~0.309 mg，0.041 6~0.624 mg，0.022 4~0.336 mg，线性关系良好(r依次为0.999 6，0.999 4，0.999 7)，平均回收率分别为94.63%，99.63%，98.11%(n=9)，RSD分别为2.1%，1.1%，1.0%。结论该方法准确可靠，重复性好，为百乐眠胶囊的质量控制提供了依据。

Abstract: OBJECTIVE To establish a method for determination of 2,3,5,4＇-tetrahydroxy stilbene-2-Ο-β-D-glucoside, polygalaxanthone III and 3,6-disinapoyl sucrose in Bai Lemian capsule by HPLC. METHODS An Agilent Eclipse XDB-C₁₈ column(4.6 mm×250 mm, 5 mm) was used. The mobile phase was methyl alcohol-acetonitrile-0.05% phosphoric acid solution (32∶8∶60), the flow rate was 1.0 mL·min^-1, and the detection wavelength was 320 nm. RESULTS The linear range for 2,3,5,4＇-tetrahydroxy stilbene-2-Ο-β-D-glucoside, polygalaxanthone Ⅲ and 3,6-disinapoyl sucrose were 0.020 6-0.309 mg, 0.041 6-0.624 mg and 0.22 4-0.336 mg(r=0.999 6, 0.999 4, 0.999 7), the average recovery were 94.63%, 99.63%, 98.11%(n=9), and RSD were 2.1%, 1.1%, 1.0%, respectively. CONCLUSION The method is accurate, credible and good reproducible. It can be used for the quality control of Bai Lemian capsule.
