黄连解毒汤主要成分的体外抗炎作用研究

    Study on in Vitro Anti-inflammatory Mechanism of the Essential Component of Huanglian Jiedu Decoction

    • 摘要: 目的 研究黄连解毒汤主要成分(黄连素、汉黄芩素)的体外抗炎作用及机制。方法 以小鼠J774A.1巨噬细胞为研究对象,分别设对照组,脂多糖(LPS)处理组,药物(不同剂量的黄连解毒汤主要成分)处理组;加入药物2 h后再加入LPS继续培养24 h,分别采用ELISA法检测关键炎症因子TNF-α、IL-6表达;逆转录聚合酶链反应(RT-PCR)检测细胞中COX-2 mRNA的水平;Western blot法测定细胞中COX-2蛋白的表达。结果 黄连解毒汤主要成分黄连素、汉黄芩素能显著抑制LPS诱导的小鼠巨噬细胞中TNF-α、IL-6的表达以及COX-2 mRNA和COX-2蛋白的表达。结论 黄连解毒汤的抗炎作用可能与其主要成分黄连素和汉黄芩素抑制TNF-α、IL-6等炎症因子的活性及下调COX-2的表达有关。

       

      Abstract: OBJECTIVE To elucidate the mechanism of anti-inflammatory effect of the essential component of Huanglian Jiedu decoction(HLJD) in vitro. METHODS RAW264·7 cells and J774A.1 cells were allocated to blank control group, model group (treated with LPS) and HLJD groups (treated with berberine, wogonin, baicalein and jasminoidin, respectively). After being treated with HLJD for 2 h and then incubated with LPS for 24 h, ELISA was used to detect the level of TNF-α, IL-6; mRNA expression of COX-2 was examined by RT-PCR, and COX-2 protein expression in cells was analysed by Western blot. RESULTS HLJD remarkably reduced TNF-α, IL-6 and COX-2 production in peritoneal macrophages elicited by LPS. CONCLUSION HLJD displays an anti-inflammatory effect mainly through inhibiting the production of inflammatory cytokines including TNF-α, IL-6 and COX-2.
