补骨脂酚在大鼠血清、肝和肾中的含量测定

Determination of Bakuchiol in Serum, Liver and Kidney of Rats

摘要: 目的建立一种快速检测大鼠灌胃给药后，补骨脂酚在血清、肝脏、肾脏中含量测定的方法。方法 Agilent HC-C₁₈柱(4.6 mm×250 mm，5 mm)；流动相：0.1%甲酸水溶液(A)-乙腈(B)，梯度洗脱(0~10 min，B 35%→45%，10~25 min，B 80%)，流速：1 mL·min^?1，柱温：30 ℃，检测波长：0~10 min，295 nm(氯霉素)，10~25 min，260 nm(补骨脂酚)。结果补骨脂酚在血清、肝匀浆液、肾匀浆液中均能达到很好的分离，在0.1~10.0 mg·L^?1内有良好的线性关系。结论本方法简便准确，可用于测定给药后血清、肝脏、肾脏中补骨脂酚的含量。

Abstract: OBJECTIVE To establish a quantitaive determination method of bakuchiol in serum, liver and kidney of rats by HPLC. METHODS The HPLC method was used with the Agilent HC-C₁₈ and the mobile phase was 0.1% formic acid-acetonitrile (65∶35) 0-10 min, B 35%→45%, 10-25 min, B 80% in the flow rate of 1 mL·min^?1. The detection wavelength was set at 295 nm(chloramphenicol) in 0-10 min and 260 nm(bakuchiol) in 10-25 min with column temperature of 30 ℃. RESULTS The bakuchiol had a good linear between 0.1-10.0 mg·L^?1 in serum, liver and kidney of rats. CONCLUSION This method can determine the content of bakuchiol in serum, liver and kidney of rats.
