HPLC测定桑叶中绿原酸、芦丁和木犀草苷含量

    Determination of Chlorogenic Acid, Rutin, and Luteolin Glycosides in Mulberry Leaves by HPLC

    • 摘要: 目的 建立同时测定桑叶中绿原酸、芦丁和木犀草苷含量的方法。方法 样品用 70%乙醇溶液提取,采用Phenomenex Luna C18色谱柱(4.6 mm×250 mm,5 μm)分离;流动相:0.1%磷酸-乙腈,梯度洗脱;流速:1.0 mL·min-1;检测波长:348 nm。结果 绿原酸、芦丁、木犀草苷分别在3.048~152.4,1.059~52.96,1.437~71.85 μg·mL-1内具有良好的线性关系;加标回收率分别为100.4%,98.3%,98.7%;RSD分别为1.7%,2.0%,1.9%(n=9)。结论 该方法简单易行,准确性和重复性好,可用于桑叶的质量控制。

       

      Abstract: OBJECTIVE To determine the content of chlorogenic acid, rutin and luteolin glycosides in mulberry leaves by HPLC. METHODS The sample was extracted by 70% ethanol and separated on a Phenomenex Luna column(4.6 mm×250 mm, 5 μm). 0.1% Phosphoric acid and acetonitrile was used as mobile phase with gradient elution at flow rate of 1.0 mL·min-1 The detection wavelength was set at 348 nm. RESULTS The linear ranges of chlorogenic acid, rutin and luteolin glycosides were 3.048-152.4, 1.059-52.96 and 1.437-71.85 μg·mL-1. And the average recoveries were 100.4%, 98.3% and 98.7% with the RSD of 1.7%, 2.0%, 1.9%(n=9), respectively. CONCLUSION This method is simple, rapid and with good repeatability, and can be used for the quality control of mulberry leaves.
