HPLC测定微球中盐酸左氧氟沙星的含量

    Determination of Levofloxacin Hydrochloride in Microspheres by HPLC

    • 摘要: 目的 建立测定壳聚糖微球中盐酸左氧氟沙星含量的高效液相色谱法。方法 采用Agilent 1200型高效液相色谱仪,色谱柱为ZORBAX Eclipse XDB-C18(4.6 mm×150 mm,5 μm),柱温:30 ℃,流速:1.0 mL·min-1,流动相:0.04 mol·L-1磷酸溶液(三乙胺调pH值为4)-乙腈(85∶15),检测波长:293 nm。结果 盐酸左氧氟沙星浓度在1.021~51.15 μg·mL-1内线性关系良好(r=0.999 6),日内和日间精密度RSD均<2%(n=5),平均加样回收率为99.1%,RSD为0.72%。结论 本方法简便易行、灵敏度高、重复性好,结果准确可靠,适用于壳聚糖微球中盐酸左氧氟沙星含量测定。

       

      Abstract: OBJECTIVE To establish an HPLC method for determination of levofloxacin hydrochloride in chitosan microspheres. METHODS The chromatographic analysis was performed on a ZORBAX Eclipse XDB-C18 column (4.6 mm×150 mm, 5 μm) at 30 ℃. The mobile phase was a mixture of 0.04 mol·L-1 phosphoric acid (adjusted to pH 4 with triethylamine) and acetonitrile (85∶15). Flow rate was 1.0 mL·min-1 and UV detection wavelength was 293 nm. RESULTS Good liner relationship was obtained in the range of 1.021-51.15 μg·mL-1(r=0.999 6). Both intra- and inter-day precision RSD were less than 2%(n=5). The average recovery rate was 99.1% with RSD of 0.72%. CONCLUSIONS The established method is sensitive, reliable and suitable for quantitative analysis of levofloxacin hydrochloride in chitosan microspheres.
