Abstract:
OBJECTIVE To establish an HPLC method for determination of eugenol in rat plasma.
METHODS HPLC was applied to assay the eugenol concentration in rat plasma after oral administration. The separation was carried out on a Allsphere ODS-2 RP-18(4.6 mm×250 mm, 5 μm) column using the mobile phase: methanol-0.5% acetic acid (63∶37), pH=4.78. The detection wavelength was set at 280 nm with the column temperature 30 ℃. The flow rate was 0.8 mL·min
-1.
RESULTS Eugenol pharmacokinetics fit the two-rooms open model after oral administration in rats with a 14.25 mg·kg
-1 dosage.
CONCLUSION The method is simple, sensitive and accurate, and could be applied to the pharmacokinetic study of eugenol in rats.
