丹参中丹参酮成分的HPLC-MS(n)研究

    Analysis of Tanshinone Constituents in Radix et Rhizoma Salviae Miltiorrhizae by HPLC-MS(n)

    • 摘要: 目的 研究丹参中丹参酮成分的ESI-MS(n)规律,建立其HPLC-MS(n)特征图谱。方法 以丹参酮ⅡA为代表,初步探讨丹参酮成分的ESI-MS(n)规律。应用HPLC-MS(n),ESI(+)模式,以水-乙腈为流动相,检测波长254 nm,梯度洗脱绘制丹参中丹参酮成分的特征图谱。结果 得出丹参酮成分的ESI-MS(n)可能的裂解途径,得到其HPLC-MS(n)特征图谱,对特征图谱中11种主要化合物进行了精确归属。结论 丹参酮类成分具有相似的ESI-MS(n)行为,根据质谱提供的各流份的分子量信息及多级质谱的裂解信息,可用于鉴别丹参酮类成分。所得到的丹参中丹参酮成分的MS(n)TIC图,可用于丹参药材中丹参酮类成分的指纹鉴别。

       

      Abstract: OBJCTIVE To search the ESI-MS(n) performance of tanshinone constituents in Radix et Rhizoma Salviae Miltiorrhizae and to establish its LC-MS(n) characteristic spectrums. METHODS Direct injection ESI-MS(n) has been used in the construction analysis of tanshinone ⅡA. Tanshinone constituents in Radix et Rhizoma Salviae Miltiorrhizae were studied by HPLC-MS(n). MS(n) was carried out on positive ion mode and HPLC was performed on a Zorbax SB-C18 column using gradient elution with a mixture of purified water and acetonitrile at the detection wavelength of 254 nm. RESULTS We got the proposed ESI-MS fragmention pathway of tanshinone constituents. By analyze of HPLC/ESI/MS(n) and HPLC/UV chromatograms, 11 characteristic peaks were determined. CONCLUSION Tanshinone constituents have similar ESI-MS(n) performance that can be used in the structure identification of tanshinone compounds. HPLC-MS (n) is a useful tool for a special identification of tanshinone constituents in Radix et Rhizoma Salviae Miltiorrhizae.
