HPLC-ELSD测定复方参芪软胶囊中黄芪甲苷的含量

Content Determination of Astragaloside in Compound Shenqi Soft Capsule by HPLC-ELSD

摘要: 目的建立以高效液相色谱-蒸发光散射检测法测定复方参芪软胶囊中黄芪甲苷含量的方法。方法色谱柱为Agilent Hypersil ODS，流动相为乙腈-水(35∶65)，流速为1.0 mL·min^-1，进样量为20 μL；蒸发光散射检测器条件：载气(压缩空气)压力为0.32 MPa，载气流量：2.6 L·min^-1，漂移管温度为108 ℃。结果黄芪甲苷进样量的线性范围为0.402 4~4.024 0 µg (r=0.999 8)；平均回收率为100.02%(RSD=1.41%)。结论该方法准确度高、专属性强，可用于该制剂中黄芪甲苷的含量测定。

Abstract: OBJECTIVE To establish an HPLC-ELSD method for the determination of astragaloside in compound shenqi soft capsule. METHODS The samples were determined by HPLC-ELSD on an Agilent Hypersil ODS column using acetonitrile-water(35∶65) as the mobile phase at a flow rate of 1.0 mL·min^-1. The injection volume was 20 μL. The ELSD conditions were as follows: pressure of compressed air was 0.32 MPa, the flow rate of compressed air was 2.6 L·min^-1，drift temperature was 108 ℃. RESULTS The linear range of astragaloside was 0.402 4-4.024 0 µg (r=0.999 8) and the average recovery rate was 100.02%(RSD=1.41%). CONCLUSION The method is accurate, specific and applicable for the determination of astragaloside in compound shenqi soft capsule.
