Abstract:
OBJECTIVE To study the technologic conditions of purifying the total flavonoids (phenolic acids) from Herba Lycopi by S-8 resin. METHODS UV-VIS spectrophotometry was used to determine the total flavonoids (phenolic acids), HPLC was used to determine caffeic acid and analyze the characteristic peaks of fingerprint. RESULTS The results showed that the effect was perfect when the concentration of original solution of the extract of Herba Lycopi was 0.1 g·mL
-1, the loading amount was 7 g Herba Lycopi per 10 g dry S-8 resin, the adsorption velocity was 1 mL·min
-1, the volume of water was 60 mL, the eluant was 80 mL 70% alcohol, and the elution velocity was 2 mL·min
-1. AB-8 resin could be used at least 5 times repeatedly after being reproducted by 95% ethanol and 1 moL·L
-1 NaOH. The remaining rate of the total flavonoids (phenolic acids) and caffeic acid was 75.56% and 104.59%, and the remaining rate of peak 1-5 was over 89%. The content of the total flavonoids and caffeic acid was raised 3.99 times and 4.69 times after being purified by the S-8 resin. CONCLUSION S-8 resin was fit for separating and purifying the total flavonoids (phenolic acids) from Herba Lycopi.
