Determination of PEG-CBT by HPLC

摘要: 目的测定聚乙二醇修饰科博肽含量。方法采用高效液相色谱法(HPLC)，色谱条件为C8色谱柱(4.6 mm×250 mm，5 μm)，0.1%三氟醋酸溶液为流动相A，三氟醋酸-80%乙腈(1∶1 000)为流动相B，流速为1 mL·min-1，检测波长为221 nm。结果该法的线性范围为51.6～258 μg·mL-1，r=0.999 4，回收率为99.9%，RSD=0.50%(n=9)。结论本法简便、准确、重复性好，可作为聚乙二醇修饰科博肽的含量测定方法。

Abstract: OBJECTIVE To determine the content of PEG-CBT. METHODS The content of PEG-CBT was determined by HPLC with C8 column (4.6 mm×250 mm, 5 μm). The mobile phase were combined with solution A (0.1% trifluoroacetic acid) and solution B (trifluoroacetic acid-80% acetonitrile (1∶1 000) . The flow rate was 1 mL·min-1, and the detection wavelength was 221 nm. RESULTS The linear range was 51.6-258 μg·mL-1 (r=0.999 4). The average recovery rate was 99.9%, and RSD was 0.50%(n=9). CONCLUSION The method is simple, accurate, and reproducible and can be used for the determination of PEG-CBT.
