Abstract:
OBJECTIVE To compare HPLC and microbiological assay for determination of the content of vancomycin in human serum. METHODS Thirty patients were given 1.0 g vancomycin by infusion,q.d..The serum concentration were determined by HPLC and bioassay,respectively. RESULTS The calibration curve of HPLC was within the range of 5.0-80.0 μg·mL-1. The recovery of HPLC method was 97.3% to 101.6%,within-day and day-to-day RSD were 1.74%-4.06%,and 1.22% -4.21%, respectively. The calibration curve of microbiological assay was linear in the range of 5.0-60.0 μg·mL-1. The recovery of microbiological assay was 98.2%-100.8%,within-day and day-to-day RSD were 4.22%-5.51% and 4.59%-6.49%,respectively. CONCLUSION The recovery and precision of the both methods are similar. There were no significant differences.
