Abstract:
OBJECTIVE To determine the contents of jateorrhizine hydrochloride, columbamine hydrochloride, epiberberine hydrochloride, coptisine hydrochloride, palmatine hydrochloride, and berberine hydrochloride in crude Coptidis Rhizoma and explore the quality relationship of Coptidis Rhizoma, which providing a basis for distinguishing.
METHODS The samples were separated by a Phenomenex Gemini-NX C
18 column (250 mm×4.6 mm, 5 μm) using acetonitrile-30 mmol·L
-1 ammonium bicarbonate (each 1 000 mL ammonium bicarbonate contained 7 mL ammonia water and 1 mL triethylamine) gradient system as mobile phase at the flow rate of 1.0 mL·min
-1, the detective wavelength was 270 nm and the column temperature was 30 ℃.
RESULTS The quality of Coptidis Rhizoma from different areas is high, the lowest average contents of jateorrhizine hydrochloride, columbamine hydrochloride, epiberberine hydrochloride, coptisine hydrochloride, palmatine hydrochloride, and berberine hydrochloride in Coptidis Rhizoma are 0.30%, 0.40%, 1.10%, 2.02%, 1.45%, 6.58%.
CONCLUSION This method is rapid, accurate, sensitive, reproducible. It is proved to be suitable for the quality control of Coptidis Rhizoma.
