基于UHPLC-Q-Exactive Focus-MS/MS分析和胃降逆胶囊的化学成分

    Analysis on Chemical Constituents of Hewei Jiangni Capsules by UHPLC-Q-Exactive Focus-MS/MS

      目的 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱法(UHPLC-Q-Exactive Focus-MS/MS)对和胃降逆胶囊中的化学成分进行定性分析。
      方法 采用Waters Acquity UPLC BEH C18色谱柱(2.1 mm×50 mm,1.7 μm),以乙腈-0.1%甲酸水为流动相进行梯度洗脱,柱温为30 ℃,流速为0.3 mL·min−1。使用电喷雾离子源,正、负离子模式扫描,扫描范围m/z 80~1 200,根据高分辨质谱数据分析和参考文献及对照品确认,对和胃降逆胶囊中的化学成分进行快速、全面的定性分析。
      结果 共鉴定出126个化合物,包括黄酮类21种、有机酸类21个、蒽醌类20个、地衣酸类16个、皂苷类13种、生物碱类4个、香豆素类4个、氨基酸类3个以及其他24个。
      结论 该化学成分研究为和胃降逆胶囊中的药效物质基础研究及质量控制提供了依据。

       

      Abstract:
      OBJECTIVE To conduct qualitative analysis of the chemical constituents in Hewei Jiangni capsules by ultra-high performance liquid chromatography coupled with quadrupole/electrostatic field orbital trap high resolution mass spectrometry(UHPLC-Q-Exactive Focus-MS/MS).
      METHODS The chromatographic separation was performed on an ACQUITY UPLC BEH C18 column(2.1 mm×50 mm, 1.7 μm) with acetonitrile-0.1% formic acid aqueous solution as mobile phase for gradient elution, the column temperature was 30 ℃ and the flow rate was 0.3 mL·min−1. Mass spectrometry was performed using an electrospray ionization source in positive and negative ion modes, with the detection range of m/z 80−1 200. The chemical constituents in Hewei Jiangni capsules were identified rapidly and comprehensively based on the accurate relative mass molecular and combined with literature data and reference substances.
      RESULTS A total of 126 chemical constituents were speculatively identified, including 21 flavonoids, 21 organic acids, 20 anthraquinones, 16 lichen acids, 13 saponins, 4 alkaloids, 4 coumarins, 3 amino acids and 24 others.
      CONCLUSION The chemical constituents study provides a basis for the research on bioactive constituents and quality control of Hewei Jiangni capsules.
