基于SIRT信号通路探讨中医药治疗溃疡性结肠炎的作用机制

    Exploring the Mechanism of Traditional Chinese Medicine in Treating Ulcerative Colitis Based on the SIRT Signaling Pathway

    • 摘要: 溃疡性结肠炎(ulcerative colitis,UC)是一种特发性的结肠黏膜慢性炎症性疾病,其发病机理较为复杂,包括肠道屏障功能受损、免疫系统调节失衡以及氧化应激反应异常等多个方面。沉默信息调节蛋白(sirtuins,SIRT)家族在UC病情进展过程中扮演着关键角色。当前UC的常规治疗药物虽能有效缓解炎症并促进黏膜愈合,但普遍存在糖皮质激素依赖、免疫抑制相关机会性感染风险升高及潜在免疫原性反应等局限性。中医药在治疗UC过程中展现出显著优势,而调节SIRT信号通路正是中医药防治UC的重要作用机制之一。本文系统概括了中药通过调控SIRT信号通路防治UC的研究进展,深入探究了SIRT激活在UC病理过程的分子机制,并阐述中药成分通过多种途径对SIRT信号通路进行调控及其潜在的作用机制,为治疗UC提供新的思路。

       

      Abstract: Ulcerative colitis(UC) is an idiopathic chronic inflammatory disease of the colonic mucosa with a complex pathogenesis involving multiple factors, including impaired intestinal barrier function, dysregulation of the immune system, and abnormal oxidative stress responses. The silent information regulator protein(sirtuin, SIRT) family plays a key role in the progression of UC. While conventional treatments for UC can effectively induce remission and maintain mucosal healing, they are associated with limitations such as corticosteroid dependence, increased risk of opportunistic infections related to immunosuppression, and potential immunogenic reactions. Traditional Chinese medicine(TCM) has demonstrated significant advantages in the treatment of UC, and regulating the SIRT signalling pathway is one of the key mechanisms by which TCM prevents and treats UC. This paper systematically reviews the research progress on TCM in preventing and treating UC by regulating the SIRT signaling pathway. It delves into the molecular mechanisms of SIRT activation in the pathological process of UC and elucidates how TCM components modulate the SIRT signaling pathway through multiple pathways and their potential mechanisms of action, providing new insights for UC treatment.
