Abstract:
OBJECTIVE To analyze chemical components of Yishen Yangyuan granules.
METHODS The ultra-highperformance liquid chromatography-quadrupole-Exactive-Plus-Orbitrap-high resolution mass spectrometry(UHPLC-Q-Exactive-Plus-Orbitrap-HRMS) technique was adopted. The separation was performed on a Hypersil GOLD C18 column with a gradient elution of 0.1% formic acid solution-acetonitrile as the mobile phase at a flow rate of 0.3 mL·min–1 and a column temperature of 35 ℃. The sample size was 1 µL. The eluent was detected in positive and negative ion modes using an electrospray ionization source. The data were processed by Compound Discoverer 3.2 and Xcalibur 4.4 software.
RESULTS A total of 237 compounds were analyzed and identified from Yishen Yangyuan granules, including 64 organic acids, 41 flavonoids, 20 terpenoids, 17 alkaloids, 16 amino acids, 11 coumarins, 12 sugars, 11 tannins, 8 stilbene glycosides, 8 anthraquinones, 5 cyclic peptides, 1 excipient, and 23 other compounds. The medicinal materials of each chemical component were attributed. Compound 17 was an organic acid, compound 151 was a flavonoid, compound 201 was a terpenoid, compound 6 was an alkaloid, and compound 179 was a coumarin. Combined with the primary and secondary mass spectrometry information, control mass spectrometry information, and literature data, the compounds were identified as malic acid, hesperidin, ajiangelin or its isomers, trigoneline and psoralen.
CONCLUSION This study analyzes and identifies the major chemical constituents of Yishen Yangyuan granules, laying the foundation for further research on the pharmacologically active substances and quality control of this formulation.
