瓜蒌薤白半夏汤化学成分的LC-MS定性定量分析

    Qualitative and Quantitative Analysis of Chemical Constituents of Gualou Xiebai Banxia Decoction Based on LC-MS

      目的  对瓜蒌薤白半夏汤的化学成分进行定性分析,并对其中9种成分进行定量测定。
      方法 采用超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(ultra-high performance liquid chromatography-quadrupole-electrostatic field orbitrap high resolution mass spectrometry,UHPLC-Q-Orbitrap-MS)对瓜蒌薤白半夏汤的化学成分进行分析,通过中药标品数据库LuMet-TCM及Herb数据库对化合物进行结构鉴定。建立高效液相色谱-串联质谱(high performance liquid chromatography-tandem mass spectrometry,HPLC-MS/MS)法同时测定该汤剂中香叶木素、知母皂苷BⅡ、琥珀酸、烟酰胺、葫芦巴碱、腺苷、5-羟甲基糠醛、鸟苷、薤白苷含量的方法,并进行仪器精密度、重复性、稳定性、线性关系、加样回收率考察。
      结果 汤剂中共鉴定出256个成分,包括氨基酸及多肽类、黄酮类、苯丙素类、生物碱类等化合物,同时定量分析瓜蒌薤白半夏汤中9种成分,其中5-羟甲基糠醛、琥珀酸、腺苷、薤白苷等含量较高,分别为540.13、521.98、25.87、23.40 μg·mL−1
      结论 本研究为瓜蒌薤白半夏汤的药效物质筛选和质量控制提供了参考。

       

      Abstract:
      OBJECTIVE To qualitatively analyze the chemical components of Gualou Xiebai Banxia Decoction and quantitatively determine the contents of 9 components.
      METHODS The chemical composition of Gualou Xiebai Banxia Decoction was analyzed using ultra-high performance liquid chromatography-quadrupole-electrostatic field orbitrap high resolution mass spectrometry(UHPLC-Q-Orbitrap-MS), and the compounds were structurally identified using the traditional Chinese medicine standard database LuMet-TCM and Herb database. A high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) was established to simultaneously determine the contents of diosmetin, timosaponin BⅡ, succinic acid, nicotinamide, trigonelline, adenosine, 5-hydroxymethylfurfural, guanosine and β-D-galactopyranoside content in this decoction. Instrumental precision, repeatability, stability, linear relationship, and sample recovery were investigated.
      RESULTS A total of 256 components were identified in the decoction, including amino acids and polypeptides, flavonoids, phenylpropanoids, alkaloids and other compounds. At the same time, 9 components in Gualou Xiebai Banxia Decoction were quantified, among which the contents of 5-hydroxymethylfurfural, succinic acid, adenosine and β-D-galactopyranoside were higher, which were 540.13, 521.98, 25.87 and 23.40 μg·mL−1, respectively.
      CONCLUSION This study provides reference for the screening and quality control of pharmacodynamic substances of Gualou Xiebai Banxia Decoction.
