类器官/器官芯片技术在生理药动学建模中的应用

    • 摘要: 传统细胞模型和动物模型在模拟人体组织的复杂性和生理相关性上有一定的局限,从而限制了临床转化效率与生理药动学模型的预测能力。类器官和器官芯片作为药物研究的新工具,更贴近体内生理过程,可模拟机体复杂的机械环境和生理反应。本文综述了类器官和器官芯片在生理药动学建模方面的研究进展,展示了这种结合在促进临床前研究向临床阶段转化并提升预测能力方面的潜力,从而为药物开发和评估提供新的视角。

       

      Abstract: Traditional cell models and animal models have certain limitations in simulating the complexity and physiological relevance of human tissues, thereby limiting both the efficiency of clinical translation and the predictive performance of physiologically based pharmacokinetic models. Organoids and organ-on-a-chip technologies, as emerging tools in drug research, more closely mimic in vivo physiological processes and can simulate the complex mechanical environments and physiological responses of the body. This article reviews the research progress of organoids and organ-on-a-chip for physiologically based pharmacokinetic modeling, demonstrating the potential of this combination to facilitate the translation from preclinical research to the clinical stage and to improve predictive capability, thereby providing new perspectives for drug development and assessment.
