基于UPLC-Q-TOF-MS的眩晕宁化学成分鉴定

    Identification on Chemical Constituents of Xuanyunning by UPLC-Q-TOF-MS

      目的 采用超高效液相色谱-四极杆飞行时间串联质谱法(UPLC-Q-TOF-MS)对眩晕宁的化学成分进行分析鉴定。
      方法 采用Waters ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),流动相为0.1%甲酸水溶液(A)-0.1%甲酸乙腈(B),梯度洗脱,流速0.50 mL·min−1,柱温40 ℃。质谱条件为选择电喷雾离子源(ESI),正、负离子检测模式,质量扫描范围50~1 500 Da。结合自建数据库、MassLynx V4.1及UNIFI软件,参考文献信息,进行眩晕宁中化学成分的鉴定。
      结果 从眩晕宁中共鉴定出包括萜类、黄酮类、苯丙素类、挥发油类、甾体类等在内的121个化学成分,其中正离子模式下46个,负离子模式下75个。
      结论 UPLC-Q-TOF-MS技术能够快速全面的分析眩晕宁的化学成分,为其后续的药效物质基础及作用机制研究奠定了基础。

       

      Abstract:
      OBJECTIVE To identify the chemical constituents of Xuanyunning by UPLC-Q-TOF-MS.
      METHODS Waters ACQUITY UPLC BEH C18 column(2.1 mm×100 mm, 1.7 μm) was employed for chromatographic separation with mobile phase of 0.1% formic acid solution(A)-0.1% formic acid-acetonitrile(B) for gradient elution, the flow rate was 0.50 mL·min−1, column temperature was 40 ℃. Electrospray ionization(ESI), positive and negative ion detection modes and mass scanning range of 50~1500 Da were selected for mass spectrometry. Combining self-built database, MassLynx V4.1 and UNIFI software, and referring to the literature information, the chemical components of Xuanyunning were identified.
      RESULTS A total of 121 components were identified in Xuanyunning, including terpenoids, flavonoids, phenylpropanoids, volatile oils, steroids and so on, of which 46 were in positive ion mode and 75 were in negative ion mode.
      CONCLUSION UPLC-Q-TOF-MS technique can be used to rapidly and sensitively identify the components of traditional Chinese medicine, and qualitative analysis of the main components of Xuanyunning can be conducted by using this technique, which provides a basis for the research of its pharmacodynamic material basis and the establishment of quality control standards.
