膜荚黄芪茎色与活性成分含量关系研究

    Study on the Relationship Between Stem Colors and Content of Active Ingredients of Astragalus Membranaceus

      目的 探究膜荚黄芪根和茎中异黄酮类、皂苷类、总多糖与黄芪茎色的关系。
      方法 优化减压内部沸腾法提取黄芪多糖工艺,采用紫外-可见分光光度法测定总多糖含量,采用UPLC-DAD法测定异黄酮类成分含量,HPLC-ELSD测定皂苷类成分含量。
      结果 黄芪多糖最佳提取工艺:解吸剂浓度为75%,解吸料液比为1∶3,解吸时间40 min,提取料液比为1∶12,提取时间45 min,提取温度80 ℃,提取次数为2次。绿茎膜荚黄芪根和茎中4种异黄酮类成分和3种皂苷类成分含量均高于紫茎膜荚黄芪,其中茎部芒柄花苷、芒柄花素、毛蕊异黄酮葡萄糖苷、黄芪甲苷含量差异极显著(P<0.01),毛蕊异黄酮含量差异显著(P<0.05),根中黄芪皂苷Ⅱ含量差异显著(P<0.05)。不同茎色黄芪根或茎部总多糖含量无统计学差异。
      结论 黄芪茎色与黄芪根、茎部异黄酮类和皂苷类活性成分含量存在一定关系,可为进一步选育优质黄芪提供理论依据。

       

      Abstract:
      OBJECTIVE  To study the relationship of isoflavones, saponins and total polysaccharides in root and stem of Astragalus membranaceus.
      METHODS  The extraction process of Astragalus polysaccharide by internal boiling under reduced pressure was optimized. Additionally, the total polysaccharide content was determined by ultraviolet-visible spectrophotometry, the isoflavone content was determined by UPLC-DAD, and the saponin content was determined by HPLC-ELSD.
      RESULTS  The optimal extraction process of Astragalus polysaccharides was as follows: the concentration of desorption agent was 75%, the solid-liquid ratio of desorption agent was 1∶3, the desorption time was 40 min, the solid-liquid ratio was 1∶12, the extraction time was 45 min, the extraction temperature was 80 ℃, and the extraction times were 2 times. The contents of 4 isoflavones and 3 saponins in the roots and stems of Astragalus membranaceus with green stem were higher than Astragalus membranaceus with purple stem. Among them, the contents of ononin, formononetin and calycosin 7-o-glucoside in stems were very significantly different(P<0.01), the contents of calycosin in stems were significantly different(P<0.05), and the contents of astragaloside Ⅱ in roots were significantly different(P<0.05). There was no significant difference in the total polysaccharide content in the roots or stems of Astragalus membranaceus with different stem colors.
      CONCLUSION  The content of isoflavones and saponins in roots and stems of Astragalus membranaceus were related to stem color, which can provide theoretical basis for further breeding and breeding of high-quality Astragalus membranaceus.
