美洲大蠊提取物对环磷酰胺所致小鼠骨髓抑制的保护作用

    Protective Effect of Periplaneta Americana Extract on Cyclophosphamide-induced Myelosuppression in Mice

      目的  观察美洲大蠊醇沉水提取物(CⅡ-3)、乙醇提取物(PA-B)对环磷酰胺(cyclophosphamide,CTX)诱导的骨髓抑制小鼠的保护作用。
      方法 小鼠腹腔注射CTX 40 mg·kg−1建立骨髓抑制模型,126只SPF级昆明种小鼠随机分为Con组,Mol组,人参皂苷Rg1组,CⅡ-3剂量组(L-CⅡ-3、M-CⅡ-3、H-CⅡ-3),PA-B剂量组(L-PA-B、M-PA-B、H-PA-B),给药治疗14 d。于第7 天、14 天尾静脉采血,测定外周血细胞。计算脾脏和胸腺指数;骨髓有核细胞(bone marrow nucleated cells,BMNC)计数;ELLSA检测促红细胞生成素(erythropoietin,EPO)、白介素-3(interleukin-3,IL-3)、白介素-6(interleukin-6,IL-6)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的表达;股骨HE染色;流式细胞术检测骨髓细胞周期和骨髓造血干细胞(hematopoietic stem cells,HSCs)含量。
      结果 第14天血细胞数值显著低于第7天,经CⅡ-3和PA-B治疗后,白细胞数目显著增加(P<0.01),脾脏指数和胸腺指数升高,BMNC数目增多(P<0.01),EPO、IL-3、IL-6、SOD表达增加,MDA表达降低(P<0.01),骨髓细胞S期和G2/M期比例升高,PI指数增加,HSCs含量增加(P<0.01)。
      结论 美洲大蠊提取物促进HSCs激活、分化和迁移,调控细胞进入S期和G2/M期,延缓骨髓细胞衰老,修复CTX造成的骨髓损伤。

       

      Abstract:
      OBJECTIVE  To observe the protective effects of alcohol-sedated aqueous extract(CⅡ-3) and ethanol extract(PA-B) of the Periplaneta americana on cyclophosphamide(CTX)-induced myelosuppression in mice.
      METHODS  CTX 40 mg·kg−1 intraperitoneal injection to establish myelosuppression model. A total of 126 SPF Kunming mice were randomly divided into Con group, Mol group, ginsenoside Rg1 group, CⅡ-3 dose group(L-CⅡ-3, M-CⅡ-3, H-CⅡ-3), and PA-B dose group(L-PA-B, M-PA-B, H-PA-B) and treated for 14 d. Blood samples were collected from the tail vein on the 7th and 14th day to measure peripheral blood cells. Spleen and thymus were taken to calculate organ index; bone marrow nucleated cell were counted; ELLSA was used to detect expression levels erythropoietin(EPO), interleukin-3(IL-3), interleukin-6(IL-6), superoxide dismutase(SOD) and malondialdehyde(MDA); HE staining of femur; flow cytometry was used to determine the bone marrow cell cycle and hematopoietic stem cells(HSCs) content.
      RESULTS  The peripheral blood cell counts were significantly lower at 14th days than at 7th days. Treatment with CII-3 and PA-B resulted in a significant increase in white blood cell counts(P<0.01), it was increased in splenic index and thymic index, increased in the number of BMNC(P<0.01), increased in levels of EPO, IL-3, IL-6 and SOD, decreased in the level of MDA(P<0.01). It increased the ratio of S and G2/M phase, increased the PI index and the content of HSCs in bone marrow(P<0.01).
      CONCLUSION  Periplaneta americana extract promotes the activation, differentiation and migration of HSCs, regulates the entry of cells into S and G2/M phases, slows down bone marrow cell senescence, and repairs bone marrow damage caused by CTX.
