佛香精油脂质体凝胶的制备及其抗痤疮丙酸杆菌作用研究

    Preparation of Citri Sarcodactylis Fructus and Caryophylli Flos Essential Oil Liposome Gel and Its Anti-propionibacterium Acnes Effect

      目的 制备佛香(佛手∶丁香)=(1∶3)精油脂质体凝胶,研究其抗痤疮丙酸杆菌(Propionibacterium acnesP. acnes)作用机制。
      方法 采用薄膜水化-高压均质法制备佛香精油脂质体,再进一步负载于卡波姆基质,即得佛香精油脂质体凝胶;利用傅里叶红外光谱、差示扫描量热仪分析精油装载状态;气相色谱-质谱联用技术分析筛选精油指标成分;改良Franz扩散池研究制剂体外经皮渗透和皮肤滞留性能;通过测定佛香精油脂质体凝胶对P. acnes抑制率、碱性磷酸酶、总蛋白及细胞膜含量研究其抑菌机制。
      结果 佛香精油装载于脂质载体疏水区内;佛香精油中指标成分为D-柠檬烯和丁香酚;佛香精油脂质体凝胶可提高精油稳定性及皮肤滞留量;佛香精油脂质体凝胶在精油含量为1.068 mg·mL−1时对P. acnes的抑制率可达到(65.9±1.7)%,可通过破坏细菌细胞壁、细胞膜结构和降低细菌蛋白质合成而抑制P. acnes生长。
      结论 佛香精油脂质体凝胶有助于提高精油脂质体稳定性且发挥缓释作用,延长药物抑菌时效,达到长效抑菌效果。

       

      Abstract:
      OBJECTIVE To prepare Citri Sarcodactylis Fructus and Caryophylli Flos(Citri Sarcodactylis Fructus∶ Caryophylli Flos)=(1∶3) essential oil liposome gel and investigate its anti-Propionibacterium acnes(P. acnes) mechanism.
      METHODS Thin film hydration-high pressure homogenization method was used to prepare Citri Sarcodactylis Fructus and Caryophylli Flosessential oil liposomes, then further loaded on carbomer matrix to obtain Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel. Fourier transform infrared spectroscopy and differentia scanning calorimetry was used to analysis the loading state of essential oil; screen index components of essential oil by gas chromatography-mass spectrometry; modified Franz diffusion cell method was used to study the transdermal penetration and dermal retention of Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel. Inhibition mechanism of Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel was investigated by detecting the inhibition rate and content of alkaline phosphatase, biofilm formation and total protein.
      RESULTS Citri Sarcodactylis Fructus and Caryophylli Flos essential oil was loaded in the hydrophobic zone of the lipid carrier. D-Limonene and eugenol were the index components of Citri Sarcodactylis Fructus and Caryophylli Flos essential oil. Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel could improve the stability and skin retention of essential oils. Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel inhibited P. acnes up to (65.9±1.7)% at essential oil level of 1.068 mg·mL−1. It could inhibit bacterial growth by destroying bacterial cell wall and membrane structure and reducing bacterial protein synthesis.
      CONCLUSION Citri Sarcodactylis Fructus and Caryophylli Flos essential oil liposomes gel helps to improve the stability of essential oils and exerts a slow-release effect, prolongs the antibacterial effect of the drug, and achieves long-term antibacterial effect.
