Abstract:
OBJECTIVE To study the therapeutic effect of Di'ao Xinxuekang on nonalcoholic fatty liver in rats.
METHODS Male SD rats were randomly divided into normal group and model group. Rats in model group were fed with high-fat and high cholesterol diet for 8 weeks. After the model was successfully established, the model rats were randomly divided into model group, atorvastatin group(2.0 mg·kg
-1) and Di'ao Xinxuekang high, medium and low(100, 30, 10 mg·kg
-1)dose groups, ten rats in each group. They were administered by gavage for 8 weeks. Oleic acid-palmitic acid(OA-PA) stimulated HepG2 cells to establish a lipid accumulation model, which was intervened by atorvastatin and Di'ao Xinxuekang. Blood lipid,liver lipid, liver function and liver index were measured. The levels of lipid and transaminase were measured. The ultrastructural changes and lipid deposition of liver and hepatocytes were observed by HE staining, transmission electron microscope and oil red O staining.
RESULTS Compared with normal group, in the model group, the liver index, blood lipid, liver lipid and liver pathological score increased significantly, liver dysfunction and the lipid accumulation and transaminase of HepG2 cells increased.Compared with model group, Di'ao Xinxuekang group and atorvastatin group decreased liver index and transaminase, significantly improved lipid metabolism and lipid accumulation, and significantly reduced hepatocyte steatosis(
P<0.05 or
P<0.01).
CONCLUSION Di'ao Xinxuekang can effectively treat nonalcoholic fatty liver in rats.
