Abstract:
OBJECTIVE To systematically evaluate the relationship between apolipoprotein E(APOE) gene polymorphism and warfarin maintenance dose in Chinese population.
METHODS PubMed, EMBase, Cochrane Library, CNKI and Wanfang databases were searched by computer to collect studies about the relationship between APOE gene polymorphisms and warfarin maintenance dose in Chinese population. The data were extracted and the quality of literature were evaluated, and meta analysis was performed by using RevMan 5.3 statistical software.
RESULTS A total of 11 studies, involving 2 320 patients were included. Meta analysis results showed that the maintenance dose of warfarin required by
APOE E2(
E2/E2, E2/E3) genotype was 30% higher than the
APOE E3(
E3/E3) genotype, and the maintenance dose of warfarin required by
APOE E2(
E2/E2, E2/E3) genotype was 43% higher than that of
APOE E4(
E3/E4, E4/E4) genotype, and the differences were statistically significant. The maintenance dose of warfarin for
APOE rs7412 TT was significantly lower than that of
CC and
CT(
P<0.05).
CONCLUSION In Chinese population, the maintenance dose of warfarin required by
APOE E2(
E2/E2, E2/E3) genotype is significantly increased, and the maintenance dose of warfarin required by
APOE rs7412 TT genotype is significantly decreased.
