复方夏天无片治疗类风湿关节炎的实验研究

    Experimental Study of Compound Xiatianwu Tablets Treatment of Rheumatoid Arthritis

    • 摘要: 目的 探讨复方夏天无片对胶原诱导性关节炎(collagen induced arthritis,CIA)小鼠的疗效及安全剂量。方法 将CIA小鼠随机分为6组:模型组、正常组、阳性(雷公藤15 mg·kg-1)组、复方夏天无片低、中、高剂量(288,864,1 728 mg·kg-1)组,每组10只。治疗21 d后,比较各组小鼠的一般情况、关节病理情况。探索复方夏天无片的安全浓度及对TNF-a诱导下的滑膜成纤维细胞(fibroblast-like synoviocyte,FLS)增殖程度的影响。结果 与模型组比较,各给药组小鼠足爪肿胀情况明显减轻,复方夏天无片中、高剂量组关节炎指数显著降低(P<0.05或P<0.01)。病理上,复方夏天无片可减少炎性细胞浸润,降低TNF-a表达,抑制破骨细胞形成,且复方夏天无片中、高剂量组优于阳性组。复方夏天无片安全浓度≤200 mg·mL-1,与模型组比较,复方夏天无片高剂量组可明显抑制FLS细胞增殖(P<0.05)。结论 复方夏天无片可改善CIA小鼠的一般情况,降低关节炎指数及TNF-a阳性表达,抑制破骨细胞形成及FLS增殖。

       

      Abstract: OBJECTIVE To explore the efficacy and safe dose of Compound Xiatianwu tablets in treating collagen-induced arthritis(CIA) of mice. METHODS CIA mice were randomly divided into 6 groups: model group, normal group, positive (Tripterygium wilfordii 15 mg·kg-1) group, Compound Xiatianwu tablets low, medium, high dose (288, 864, 1 728 mg·kg-1) groups, with 10 mice in each group. After 21 d of treatment, the general condition and joint pathology were compared wtih each group, and the safe concentration of Compound Xiatianwu tablets and its effect on proliferation of TNF-α induced fibroblast-like synoviocyte(FLS) were explored. RESULTS Compared with model group, paw swelling of mice in treatment groups was all reduced, and arthritis index of mice in Compound Xiatianwu tablets medium, high dose groups was significantly decreased(P<0.05 or P<0.01). Pathologically, Compound Xiatianwu tablets could reduce inflammatory cells infiltration, decrease the positive expression of TNF-α and inhibit the formation of osteoclasts, and Compound Xiatianwu tablets medium, high dose groups had better effects than positive group. The safe concentration of Compound Xiatianwu tablets was ≤ 200 mg·mL-1. Compared with model group, Compound Xiatianwu tablets high dose group had a good inhibitory effect on proliferation of FLS and there were statistically significant differences(P<0.05). CONCLUSION Compound Xiatianwu tablets can improve the general conditions of CIA mice, reduce the arthritis index, decrease the positive expression of TNF-α, inhibite osteoclast formation and FLS proliferation.
