槐白皮提取物对CT26荷瘤小鼠的抑瘤减毒作用研究

    Study on the Anti-tumor and Toxicity Reduction Effects of the Bark Extract of Sophora Japonica on the CT26 Tumor-bearing Mice

    • 摘要: 目的 探讨槐白皮提取物对CT26荷瘤小鼠的抑瘤减毒作用。方法 BALB/c小鼠建立CT26荷瘤小鼠模型,建模成功后分为生理盐水组、氟尿嘧啶(5-FU)组、联合用药组和槐白皮提取物组,连续给药9 d,给药过程中检测小鼠体质量变化,给药结束后计算抑瘤率和免疫器官系数,HE染色观察肿瘤和肝脏组织形态学变化,ELISA检测血清白细胞介素-2(IL-2)和肿瘤坏死因子-α(TNF-α)水平,免疫组化检测肿瘤组织中Ki-67蛋白的表达水平。结果 5-FU组、联合用药组和槐白皮提取物组抑瘤率分别为77.02%,81.75%,46.62%。与生理盐水组比较,槐白皮提取物组瘤重明显降低,槐白皮提取物组脏器系数、血清IL-2和TNF-α水平明显升高(P < 0.05或P < 0.01),HE染色结果显示槐白皮提取物组肿瘤组织坏死区增多,免疫组化结果显示槐白皮提取物组肿瘤组织Ki-67阳性细胞数减少。与5-FU组比较,联合用药组小鼠体质量下降程度和肝细胞水肿程度降低。结论 槐白皮提取物对CT26荷瘤小鼠有抑瘤作用,并且对5-FU化疗引起的小鼠体质量降低和肝细胞损伤有保护作用。

       

      Abstract: OBJECTIVE To explore the the anti-tumor and toxicity reduction effects of the bark extract of Sophora japonica on the CT26 tumor-bearing mice. METHODS BALB/c mice were used to establish CT26 tumor-bearing mice model. The model mice were divided into normal saline group, 5-fluorouracil(5-FU) group, combination group and the bark extract of Sophora japonica group. All groups were administered for nine days. During administration, change of mice's body weight were detected. After the end of administration, the tumor inhibition rate and the immune organ index was measured. Morphological changes in tumor and liver tissues were observed by HE staining. The levels of interleukin-2(IL-2) and tumor necrosis factor-α(TNF-α) in serum were measured by ELISA. The expression levels of Ki-67 in tumor tissues were detected by immunohistochemistry. RESULTS The tumor inhibition rate of 5-FU group, combination group and the bark extract of Sophora japonica group were 77.02%, 81.75%, 46.62%, respectively. Compared with normal saline group, the bark extract of Sophora japonica group significantly decreased the tumor weight, and significantly increased the immune organ index, IL-2 and TNF-α levels in serum(P < 0.05 or P < 0.01). The results of HE staining showed that the necrotic areas of tumor tissues in the the bark extract of Sophora japonica group were significantly increased. Immunohistochemical results showed that the number of Ki-67 positive cells was obviously decreased in the bark extract of Sophora japonica group. Compared with the 5-FU group, the weight loss and hepatocyte edema in combination group were reduced. CONCLUSION The bark extract of Sophora japonica possesses anticancer effect on CT26 tumor-bearing mice and has protective effect on weight loss and hepatocyte injury of mice induced by 5-FU chemotherapy.
