基于ICP-MS研究断体地龙对大鼠创面无机元素含量的影响

    Study on effect of tail-amputated earthworm on Contents of inorganic elements in wound Skin of rats by ICP-MS

    • 摘要: 目的 建立微波消解-ICP-MS测定大鼠创面皮肤中无机元素含量,考察断体地龙促创面愈合作用与无机元素的关系。方法 采用HNO3体系-微波消解法对样品进行前处理,ICP-MS对大鼠创面皮肤中钾、镁、钙、锰、铁、钴、镍、铜、锌、砷、硒、镉、碲、铅14种无机元素进行测定和分析。结果 在优化的实验条件下,元素在0~250 μg·L-1内回归方程系数r ≥ 0.999;方法的检出限(3σn=10)为0.02~13.04 μg·L-1;加样回收率(n=6)为85.4%~119.0%。断体地龙可明显提高创面组织中钾、钙、镁、铜、铁、锌、硒的含量,且未增加重金属元素的含量。结论 该方法简便快速、灵敏度高、准确性好,可用于同时测定大鼠创面皮肤中多种无机元素的含量。断体地龙发挥促创面愈合作用与提高各无机元素含量有关,且具有较高的安全性。

       

      Abstract: OBJECTIVE To establish method for the content determination of inorganic elements in wound skin of rats by ICP-MS with microwave digestion, and to investigate the relationship between the effect of tail-amputated earthworm on wound healing and inorganic elements. METHODS The samples were pre-treated by HNO3 system-microwave digestion method, and 14 inorganic elements including K, Mg, Ca, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Cd, Te and Pb were determined and analyzed by ICP-MS. RESULTS The method showed good linearity at 0-250 μg·L-1(r ≥ 0.999) under optimized experimental conditions. The limit of detection (3σ, n=10) was 0.02-13.04 μg·L-1. The recovery(n=6) was 85.4%-119.0%. The content of K, Ca, Mg, Cu, Fe, Zn and Se in the wound tissue was obviously increased by tail-amputated without increasing the content of heavy metal elements. CONCLUSION This method is simple, rapid, sensitive and accurate, and it can be used for simultaneous determination of inorganic elements on wound skin of rats. Tail-amputated earthworm plays a role in promoting wound healing, which is related to increasing the content of inorganic elements and has a higher safety.
