HPLC测定人工关节假体感染患者关节液中万古霉素浓度

    Determination of Vancomycin Concentration in Synovial Fluid of Patients with Prosthetic Joint Infection by HPLC

    • 摘要: 目的 建立HPLC测定人工关节假体周围感染患者关节液中万古霉素浓度的方法。方法 以替硝唑为内标,采用Hypersil C18柱(250 mm×4.6 mm,5 μm),流动相为乙腈-0.012 5mol·L-1磷酸二氢钾缓冲液(pH=3.2)(10:90),流速1 mL·min-1,柱温35℃,检测波长236 nm,进样量10 μL。结果 关节液中万古霉素浓度在0.906 8~90.681 8 μg·mL-1线性良好。批内、批间精密度RSD均<15%,准确度85.89%~103.51%。结论 本法易于操作,精密度和准确度良好,适用于人工关节假体周围感染患者关节液中的万古霉素浓度监测。

       

      Abstract: OBJECTIVE To establish a method for determination of vancomycin concentration in synovial fluid of patients with prosthetic joint infection by HPLC. METHODS Used tinidazole as internal standard substance, vancomycin concentration in synovial fluid was analyzed by Hypersil C18(250 mm×4.6 mm, 5 μm) with mobile phase of acetonitrile-0.012 5 mol·L-1 potassium dihydrogen phosphate buffer solution(pH=3.2)(10:90). The flow rate was 1 mL·min-1. The column temperature was 35℃ and the injection volume was 10 μL. RESULTS Vancomycin concentration in synovial fluid was in well linearity in the range of 0.906 8-90.681 8 μg·mL-1. The relative standard deviations of intra-batch and inter-batch precision was <15%, and the accuracy was between 85.89%-103.51%. CONCLUSION The method is simple, sensitive and accurate, which is suitable for monitoring of vancomycin concentration in synovial fluid of patients with prosthetic joint infection.
