Abstract:
OBJECTIVE To investigate the effect of scutellarin on JAK/STAT signaling pathway and its apoptotic mechanism in non-small cell lung cancer cells.
METHODS Non-small cell lung cancer A549 cells were cultured and collected cells after treatment with different concentrations of scutellarin for 24 h. Cell proliferation was analyzed by MTT method, apoptosis was evaluated by flow cytometry. The mRNA expression of JAK2, STAT3, Pim1 and Bcl-2 were detected by RT-PCR; JAK2 and STAT3 protein were detected by Western blot.
RESULTS MTT assay and flow cytometry showed that scutellarin reduced the survival rate of A549 cells and promoted its apoptosis in a dose-dependent manner. RT-qPCR and Western blot showed that compared with the control group, the mRNA expression of JAK2, STAT3, Pim1 and Bcl-2 were decreased in the scutellarin group, JAK2 and STAT3 protein were decreased, which were inversely proportional to the concentration of scutellarin.
CONCLUSION Scutellarin can promote apoptosis of non-small cell lung cancer A549 cells, which may be through inhibition of JAK/STAT signaling pathway activation and reduction of JAK2 and STAT3 protein expression.
