Abstract:
OBJECTIVE To compare two methods for preparing mouse models of acute gouty arthritis.
METHODS Eighteen C57BL/6 male mice were divided into 3 groups randomly, building acute gouty arthritis model mice with 25 μL 50 mg·mL
-1 monosodium urate injection into ankle joint cavity and toe pad respectively, blank group using 0.9% NaCl instead. Weight and foot swelling were compared, as well as expression of IL-1β in ankle fluid and serum. Furthermore, observed the histomorphology in each group.
RESULTS There was no statistical difference in weight before or after modeling. The highest expression of IL-1β was found in the articular fluid of the ankle joint group, which was statically significant compared with the foot pad group and blank group(
P<0.05 or
P<0.01). The intensity of inflammatory cell infiltration was ankle joint group > toe pad group > blank group.
CONCLUSION Using ankle joint modeling method and taking ankle joint fluid as ELISA sample is conducive to the animal experimental study of acute gouty arthritis.
