Jatamanvaltrate P对乳腺癌细胞T47D增殖的影响及机制研究

    • 摘要: 目的 考察蜘蛛香中环烯醚萜类化合物Jatamanvaltrate P对乳腺癌细胞T47D的增殖影响及作用机制。方法 MTT法检测Jatamanvaltrate P对乳腺癌细胞T47D和BT-20增殖的影响;DAPI染色和流式细胞术检测Jatamanvaltrate P对T47D细胞凋亡和周期的影响;Western blot检测与细胞周期、凋亡相关蛋白的变化。结果 Jatamanvaltrate P能明显抑制乳腺癌细胞增殖,并且呈时间-剂量依赖性;Jatamanvaltrate P能明显将T47D细胞阻滞于G2/M期,下调周期相关蛋白cdc-2和Cyclin B1,上调p-cdc-2;Jatamanvaltrate P能诱导T47D细胞产生凋亡小体,显著性增加T47D细胞凋亡比例,呈现剂量依赖性;Jatamanvaltrate P处理后的蛋白PARP和Caspase 3/8/9出现了活化形式。结论 Jatamanvaltrate P通过阻滞细胞周期、诱导细胞凋亡来抑制乳腺癌细胞T47D的增殖,有望成为一种潜在的治疗乳腺癌的药物。

       

      Abstract: OBJECTIVE To investigate the effect and mechanism of Valeriana jatamansi Jones. extract Jatamanvaltrate P on proliferation of T47D cells to breast cancer. METHODS MTT assay was used to detect the effect of Jatamanvaltrate P on T47D and BT-20 cells; DAPI staining and flow cytometry(FCM) were used to examine cell cycle and apoptosis; Western blot assay was performed to detect the expression of related proteins of cell cycle and apoptosis. RESULTS Jatamanvaltrate P significantly inhibited the breast cancer cells in a time-and dose-dependent; Jatamanvaltrate P induced cell cycle arrest at G2/M phase, and the expression of cycle associated proteins Cyclin B1 and cdc2 was decreased, while p-cdc2 was increased; Jatamanvaltrate P was able to induce T47D cells to produce apoptotic bodies and significantly increase the apoptosis rate of T47D cells in a dose-dependent manner. The proteins of PARP, Caspase 3, Caspase 8 and Caspase 9 produced activated form in T47D cells by treatment with Jatamanvaltrate P. CONCLUSION Jatamanvaltrate P may be possible to inhibit the proliferation of T47D cells by cell cycle arrest and inducing apoptosis, which is expected to become a potential drug for the treatment of breast cancer.
