Abstract:
OBJECTIVE To evaluate the effect of different formulation (aerosol and oral) of Chinese classic formula Danggui-beimu-kushen on chronic obstructive pulmonary (COPD) murine model and to study its mechanisms.
METHODS The bronchoalveolar lavage fluid (BALF) and lung tissues were collected.Differential WBC count and content of cytokine (KC,MIP2,TNF-
α) in BALF,HE staining for lung biopsy,KC and Muc5AC mRNA expression of lung and content in BALF were determined.
RESULTS Both the gavage and aerosol groups decreased infiltration of the inflammatory cells than that of the control group,moreover,the aerosol group showed better effect than that of gavage group.The gavage and aerosol groups could further decrease the content of KC,MIP-2 and mRNA expression of Muc5AC.In addition,aerosol group could substantially suppress the content of TNF-a and inhibited mRNA expression of KC.
CONCLUSION Aerosol formula showed better effect on the pulmonary inflammation and mucus secretion in COPD murine model than that of gavage formula.The mechanism were involved in decreasing TNF-
α content,and mRNA expression of KC.
