基于caspase-3/bcl-2/bax信号通路的槐定碱诱导胰腺癌细胞株capan-1凋亡机制研究

    Influence of Sophoridine on Proliferation and Caspase-3/bcl-2/bax Signaling Pathway of Human Pancreatic Cancer Cell Line Capan-1 Cells

    • 摘要: 目的 探讨槐定碱对人胰腺癌细胞株capan-1作用及机制。方法 MTT法检测细胞增殖;Hoechst33342染色法观察细胞凋亡;流式检测细胞凋亡和细胞周期百分率;Western blot法检测caspase-3/bcl-2/bax表达。结果 槐定碱可以成浓度依赖性诱导capan-1细胞增殖抑制和凋亡,增加S/G2期细胞比例,下调bcl-2和pro-caspase-3表达水平,上调bax蛋白的表达。结论 槐定碱能通过调节caspase-3/bcl-2/bax信号通路而诱导细胞凋亡。

       

      Abstract: OBJECTIVE To explore the effects and mechanisms of sophoridine on human pancreatic cancer cell line capan-1. METHODS MTT, Hoechst33342 staining and flow cytometric analysis were used to evaluate the proliferation, apoptosis and cell cycle percentage. Western-blot assay were used to evaluate expression of caspase-3/bcl-2/bax protein. RESULTS Sophoridine could induce the proliferation inhibition, apoptosis and the expression of bcl-2 and pro-caspase-3 with increasing S/G2 percentage and the expression of bax on capan-1 cell in a concentration dependent manner. CONCLUSION Sophoridine can induce the apoptosis of capan-1 cell by regulating caspase-3/bcl-2/bax signaling pathway.
