罗红霉素的水杨基荧光酮荷移分光光度法测定

摘要:
目的测定罗红霉素胶囊中罗红霉素的含量,增加质量控制和保证方法。方法利用罗红霉素与水杨基荧光酮(SAF)形成1∶1荷移络合物,应用分光光度法测定胶囊中罗红霉素含量。结果络合物的最大吸收波长是516 nm,表观摩尔吸光系数ε为5.01×103 L·mol-1·cm-1,罗红霉素在33.5~217.6 mg·L-1内符合比尔定律,RSD为0.64%(n=8),平均回收率为100.9%。结论方法简单、灵敏、准确,可作为该药的含量测定方法。

Abstract:
OBJECTIVE To determine roxithromycin in capsules and provide the quality control and assurance. METHODS A spectrophotometric method based on the charge-transfer reaction between salicyl fluorone (SAF) and roxithromycin. RESULTS The molar absorptivity of the complex at 516 nm was 5.01×103 L·mol-1·cm-1. Beer's law was obeyed in the range of 33.5-217.6 mg L-1of roxithromycin. The relative standard deviation was 0.64%(n=8). The composition of the complex was found to be 1∶1 by slope ratio and Job’s methods. The mechanism of reaction was investigated, and the average recovery was found to be 100.9%. CONCLUSION The method is simple, quick and accurate for the determination of roxithromycin in capsules.
