葛根素对高脂血症大鼠血脂、血凝及血小板聚集的影响

摘要:
目的 研究葛根素对高脂血症大鼠血脂、血凝及血小板聚集的影响。方法 建立大鼠的高脂血症模型,同时连续腹腔注射给予葛根素(50 mg·kg-1)或空白溶媒(0.9% NaCl)30 d,测定大鼠血浆总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、丙二醛(MDA)、超氧化物歧化酶(SOD)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、ADP诱导的血小板5 min最大聚集率(MAR)。结果 与高脂饲料组比较,饲高脂饲料大鼠在连续腹腔注射给予葛根素(50 mg·kg-1)30 d后,大鼠血浆TG含量平均下降27%,TC含量平均下降21%,LDL-C含量平均下降39%,HDL-C含量平均增加24%;MDA降低,SOD升高,血浆PT、APTT显著延长、MAR降低(P<0.05)。结论 葛根素不但能有效降低高脂血症大鼠血脂水平,而且可能通过防止脂质过氧化而有效改善动物血凝系统。

Abstract:
OBJECTIVE To explore effects of puerarin on lipidemia, blood coagulation and platelet aggregation in experimental hyperlipidemia rats. METHODS Experimental hyperlipidemia rats were ip administration of puerarin (50 mg·kg-1·d-1) or vehicle (0.9% NaCl) in 30 d. The levels of plasma triplyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), malondialdehyde (MDA), superoxide dismutase (SOD), prothrombin time (PT), activated partial thrombplastin time (APTT), and platelet maximal aggregation rate(MAR) which was induced by ADP in 5 min were measured. RESULTS Plasma TG, TC, LDL-C and HDL-C of the rats fed high fat diet (HFD) and ip puerarin (50 mg·kg-1·d-1) were 27%, 21%, 39% lower and 24% higher than that of HFD group rats. The PT and APTT of puerarin and HFD group rats were significantly longer than that of the HFD group (P<0.05). The SOD in puerarin and HFD group was higher (P<0.05) and the MDA and MAR in puerarin and HFD group was lower than that of HFD group (P<0.01). CONCLUSION Puerarin can reduce the serum lipid level of the hyperlipidemia rats effectively and prevent the lipid peroxidation, moreover blood clotting system in hyperlipidemia rats is improved effectively.
