OBJECTIVE To conduct qualitative analysis of the chemical constituents in Hewei Jiangni capsules by ultra-high performance liquid chromatography coupled with quadrupole/electrostatic field orbital trap high resolution mass spectrometry(UHPLC-Q-Exactive Focus-MS/MS).
METHODS The chromatographic separation was performed on an ACQUITY UPLC BEH C18 column(2.1 mm×50 mm, 1.7 μm) with acetonitrile-0.1% formic acid aqueous solution as mobile phase for gradient elution, the column temperature was 30 ℃ and the flow rate was 0.3 mL·min−1. Mass spectrometry was performed using an electrospray ionization source in positive and negative ion modes, with the detection range of m/z 80−1 200. The chemical constituents in Hewei Jiangni capsules were identified rapidly and comprehensively based on the accurate relative mass molecular and combined with literature data and reference substances.
RESULTS A total of 126 chemical constituents were speculatively identified, including 21 flavonoids, 21 organic acids, 20 anthraquinones, 16 lichen acids, 13 saponins, 4 alkaloids, 4 coumarins, 3 amino acids and 24 others.
CONCLUSION The chemical constituents study provides a basis for the research on bioactive constituents and quality control of Hewei Jiangni capsules.
