OBJECTIVE To develop an efficient chemical method for synthesizing chitosan tetrasaccharide, thereby providing a material basis for investigating the structure-activity relationship of chitooligosaccharides.
METHODS Utilizing D-glucose as the starting material, with rationally designed orthogonal protection strategies, the synthetic route was optimized to enhance the synthesis efficiency of chitosan tetrasaccharide.
RESULTS A highly efficient synthetic route was successfully established, requiring only 12 steps to complete the preparation of key glycosyl donors and acceptors, achieving the precise synthesis of the target tetrasaccharide.
CONCLUSION This study presents an efficient synthesis strategy for the synthesis of a chitosan tetrasaccharide, featuring a minimal use of protecting groups and building blocks, as well as streamlined synthetic steps, thereby demonstrating strong potential for scalable production.
