体位性低血压大鼠模型的建立及应用研究

    Establishment and Application of A Rat Model of Orthostatic Hypotension

    • 摘要: 目的 建立体位性低血压大鼠模型并进行初步应用。方法 从倾斜角度(30°,45°,75°,90°)和倾斜时间(1,2 min)两方面优化建立大鼠低血压模型的条件,再用哌唑嗪(1.1~220 μg·kg-1)和萘哌地尔(30~3 000 μg·kg-1)多剂量给药,探讨模型有效性和可靠性。结果 随着倾斜角度的增加,大鼠的血压和心率变化幅度加大,体位持续时间与血压、心率的变化没有明显的相关性。哌唑嗪剂量依赖性强化了体位性低血压,对血压和心率都表现出明显的抑制作用;萘哌地尔仅在3 000 μg·kg-1剂量表现出对血压和心率的抑制效应。结论 45°头朝上位倾斜、持续时间60 s是建立体位性低血压大鼠模型的最适条件,哌唑嗪和萘哌地尔检验的结果与文献报道一致,证明了本模型有效、可靠。

       

      Abstract: OBJECTIVE To explore establishmentand preliminary application of a rat model of orthostatic hypotension. METHODS The rat model of orthostatic hypotension was optimized from angle of tilt(30°, 45°, 75°, 90°)and tilt time(1 min or 2 min). The validity and reliability of the model were observed by administrated with prazosin(1.1-220 μg·kg-1) and naphthalene (30-3 000 μg·kg-1) RESULTS With the increase of angle, variation amplitude of rats’ blood pressure and heart rate all increased. The duration of position and the change of blood pressure and heart rate had no obvious correlation. Prazosin dose dependent strengthened orthostatic hypotension. Blood pressure and heart rate were obviously inhibited. Only 3 000 μg·kg-1 naphthalene had inhibitory effect on blood pressure and heart rate. CONCLUSION 45°head-up tilt and 60 s is the optimum condition to establish a rat model of orthostatic hypotension; the results of administrating prazosin and naphthalene are consistent with the literature reports proving the validity and reliability of the model.
