HPLC测定瑞替加滨的含量

    Determination of Retigabine by HPLC

    • 摘要: 目的 建立HPLC测定瑞替加滨含量的方法。方法 色谱柱为Agilent ZORBAX SB-C18(150 mm×4.6 mm,3.5 μm),流动相为庚烷磺酸钠溶液-乙腈,采用梯度洗脱,流速为1.0 mL·min-1,检测波长为250 nm,温度为30 ℃。结果 瑞替加滨在0.204~122.28 μg·mL-1内,峰面积与浓度呈良好的线性关系(r=0.999 9),平均回收率99.9%,RSD=0.46%。结论 本法准确、重复性好,能与其同分异构体较好分离,适用于瑞替加滨的含量测定。

       

      Abstract: OBJECTIVE To establish a method to determine the content of retigabine. METHODS Agilent ZORBAX SB-C18(150 mm×4.6 mm, 3.5 μm) was used as the analytical column. The mobile phase was a mixture of sodium heptanesulfonate solution-acetonitrile, and elute with linear gradient. The flow rate was 1.0 mL·min-1. Detection wavelength was 250 nm and column temperature was 30 ℃. RESULTS Retigabine has a good linearity in the range of 0.204-122.28 μg·mL-1 (r=0.999 9), the average recovery was 99.9% with RSD of 0.46%. CONCLUSION This method is accurate, repeatable and its isomers can be well separated. It can be applied to determine the content of retigabine.
