Abstract:
OBJECTIVE To establish a method for the detection of encapsulation rate of gatifloxacin in gatifloxacin-polylacticacid nanoparticles. METHODS The analysis was performed on a sephadex G-50 column, using distilled water as eluent, and detected by HPLC with a C
18 column (150 mm×4.6 mm, 5 μm) using 0.02 mol·L
-1 citric acid (0.6% triethylamine)-methanol (60∶40) as mobile phase under the flow rate of 1.0 mL·min
-1. The detection wavelength was 290 nm. RESULTS Gatifloxacin had a good linear range of 2.051-60.14 μg·mL
-1(
r=0.999 9). The average column recovery rate was 98.8% with RSD of 0.71%. The average encapsulation rate of 3 batchs nanoparticles was 82.1% with RSD of 2.2%. CONCLUSION The method can be used to detect the encapsulation rate of gatifloxacin in the nanoparticles.
