Abstract:
OBJECTIVE To prepare fluorescent probe coumarin-6 loaded nanoparticles using the biodegradable material poly(lactic-co-glycolic acid)(PLGA) as carrier.
METHODS Coumarin-6 loaded PLGA nanoparticles were prepared by emulsification solvent evaporation method. The formulation was optimized by orthogonal design using utilization ratio and entrapment efficiency as the indexes.
RESULTS The mean entrapment efficiency of coumarin-6 loaded PLGA nanoparticles was 51.6% and the utilization ratio was 81.9%. The average size was 135 nm and the
in vitro cumulative release of coumarin-6 from nanoparticles was less than 2% in 72 h.
CONCLUSION The preparation process of coumarin-6 loaded PLGA nanoparticles is simple and feasible, and coumarin-6 can be used as fluorescence marker for in vivo tracing and targeting study of nanoparticles.
