基于UPLC-Q-TOF-MS/MS技术比较桂枝-甘草药对单煎液和合煎液的入血成分

    Comparison of Blood Components in Single and Combined Decoction of Cinnamomi Ramulus-Glycyrrhizae Radix et Rhizoma Medicinal Pairs Based on UPLC-Q-TOF-MS/MS

      目的  对桂枝-甘草药对单煎液和合煎液的入血成分进行分析比较,为桂枝-甘草药对的药效物质基础研究提供参考。
      方法 通过UPLC-Q-TOF-MS/MS技术,根据提取离子流色谱图、质谱碎片裂解信息识别目标化合物,结合TCMSP、BATMAN-TCM以及知网等综合性数据库,筛选入血成分。对桂枝、甘草以及药对含药血清的化学成分进行分析。
      结果 桂枝、甘草及其药对均含有丰富的化学成分。从桂枝、甘草及其药对含药血清中分别鉴定出103个、164个、143个化学成分。其中桂枝入血原型成分20个,代谢产物83个;甘草含药血清入血原型成分41个,潜在入血原型成分代谢产物123个;桂枝-甘草药对含药血清入血原型成分31个,代谢产物112个;主要入血成分包括黄酮类、酚类、苯丙素类及萜类。
      结论 本研究对桂枝-甘草单煎液和合煎液入血成分进行了定性分析,明确了其入血成分的差异。为桂枝-甘草药对的药效物质基础和作用机制的进一步研究提供参考。

       

      Abstract:
      OBJECTIVE  To analyze and compare the blood-entry components of single decoction and combined decoction of Cinnamomi Ramulus, Glycyrrhizae Radix et Rhizoma, and their medicinal pairs, so as to provide a reference for the basic research on the pharmacological substances.
      METHODS  The target compounds were identified by UPLC-Q-TOF-MS/MS technique based on extracted ion flow chromatograms, mass spectrometry fragmentation cleavage information, and screened for haematological constituents by combining comprehensive databases such as TCMSP, BATMAN-TCM, and CNKI Networks. The chemical composition of Cinnamomi Ramulus-Glycyrrhizae Radix et Rhizoma medicinal pair containing serum was analysed.
      RESULTS The blood-entry components of Cinnamomi Ramulus, Glycyrrhizae Radix et Rhizoma, and their medicinal pairs were rich in chemical constituents. Total 103, 164, 143 constituents were identified from the blood serum of Cinnamomi Ramulus, Glycyrrhizae Radix et Rhizoma, and their medicinal pairs, respectively. Cinnamomi Ramulus plasma samples containing 20 potential prototype blood-entry components, and 83 metabolites of potential prototype blood-entry components.Glycyrrhizae Radix et Rhizoma plasma samples containing 41 potential prototype blood-entry components, and 123 metabolites of potential prototype blood-entry components. Cinnamomi Ramulus-Glycyrrhizae Radix et Rhizoma medicinal pairs plasma samples containing 31 potential prototype blood-entry components, and 112 metabolites of potential prototype blood-entry components. Mainly differentiated including flavonoids, phenolics, phenylpropanoids, alkaloids, and other components.
      CONCLUSION  In this study, the blood-entry components of Cinnamomi Ramulus-Glycyrrhizae Radix et Rhizoma single decoction and combined decoction are qualitatively analysed, and the differences in blood-entry components are clarified. It provides a reference for further studies on the pharmacological material basis and mechanism of action of Cinnamomi Ramulus-Glycyrrhizae Radix et Rhizoma medicinal pairs.
