Abstract:
OBJECTIVE To analyze the main components of Baidoukou aqueous extract and the components absorbed into blood of rats after oral administration by UPLC-Q-Exactive-Orbitrap MS/MS technology.
METHODS Using ACQUITY UPLC HSS T3(2.1 mm×100 mm, 1.8 μm) column; using A(deionized water, containing 0.1% formic acid) and B(acetonitrile, containing 0.1% formic acid) as mobile phases with gradient elution, column temperature was 35°C, while injection volume was 10 μL, and flow rate was 0.2 mL·min−1. Using electrospray ionization source(ESI), detect with Full MS/dd-MS2 mode.
RESULTS A total of 70 components were identified from the aqueous extract of Baidoukou, including organic acids, flavonoids, amino acids, alkaloids, and lactones. Nineteen prototype components were identified in the drug-containing serum samples, including 10 organic acids such as artemisinic acid and azelaic acid, 2 lactones such as isoalantolactone and dehydrocostus lactone, 2 alkaloids such as betaine and xanthine, 3 amino acids such as L-tyrosine and L-tryptophan.
CONCLUSION This study sclarified the chemical composition of Baidoukou aqueous extract and its oral blood components for the first time, providing a theoretical basis for its further research and development.
