响应曲面分析法研究5-FU温热化疗对BGC-823细胞增殖的影响

摘要:
目的 采用响应曲面分析法研究5-FU温热化疗对BGC-823细胞增殖的影响。方法 取对数生长期的BGC-823细胞,考察5-FU不同孵育时间、不同孵育温度、不同药物浓度对BGC-823细胞抑制率的影响,运用Design-export 8.5软件对各指标数值进行整合,并构建三维响应曲面图进行交互作用分析,优化出最佳的温热化疗条件,并进行验证试验。结果 经过整合分析发现,5-FU药物浓度、孵育时间和孵育温度3个因素对BGC-823细胞增殖均有影响,其影响的排序为药物浓度 > 孵育时间 > 孵育温度;另外,孵育时间与孵育温度之间的交互作用也很显著。优化后的温热化疗条件:药物浓度16 000 μg·mL-1、孵育时间20 min、孵育温度40℃,经3次验证试验,证实细胞抑制率可以达到43.18%,比模型预测值偏差仅0.85%。结论 温热化疗方法可以增强5-FU对BGC-823细胞的抑制作用。

Effects of 5-FU Thermochemotherapy on the Proliferation of BGC-823 Cells by Response Surface Methodology

Abstract:
OBJECTIVE To study the effect of 5-FU thermochemotherapy on the proliferation of BGC-823 cells by response surface methodology. METHODS The logarithmic growth of BGC-823 cells was used to investigate the effects of 5-FU on the inhibition rate of BGC-823 cells, including:incubation time, incubation temperature and drug concentration. Design-export 8.5 was used to integrate the values of each index, and a three-dimensional response surface graph was constructed to analyze the interaction. The optimal thermochemotherapy conditions were optimized, and the optimized experiment conditions were verified. RESULTS After integrated analysis, it was found that the concentration, incubation time and incubation temperature all had effects on the proliferation of BGC-823 cells. The order of effects was drug concentration > incubation time > incubation temperature. In addition, the interaction between incubation time and incubation temperature was also significant. The optimized thermochemotherapy conditions were:16 000 μg·mL-1 of drug concentration, 20 min of incubation time, 40℃ of incubation temperature. After three validation tests, it was confirmed that the cell inhibition rate could reach 43.18%, which was only 0.85% deviation from the predicted value of the model. CONCLUSION Thermotherapy can enhance the inhibitory effect of 5-FU on BGC-823 cells.
