Abstract
OBJECTIVE To establish a method for the simultaneous determination of hydroxysaffor yellow A(HSYA) and chlorogenic acid in Fufang Honghua spray. METHODS The dual-wavelength HPLC was adopted. The Shim-pack VP-ODS column(250 mm×4.6 mm, 5 μm) was used with mobile phase of acetonitrile-0.4% phosphonic acid(12∶88) at flow rate of 1.0 mL·min-1, and the detection wavelength was set at 403 nm for HSYA and 327 nm for chlorogenic acid. The column temperature was 30 ℃. RESULTS The method displayed good linearity over the concentration ranges of 0.612-12.24 μg·mL-1 for HSYA and 0.632-12.64 μg·mL-1 for chlorogenic acid(r=0.999 9), respectively. The average recoveries were 98.34% and 99.82%, with the RSD of 1.16% and 1.64% (n=9), respectively. CONCLUSION This method is simple, rapid, reliable and reproducible. It can be used for determination of HSYA and chlorogenic acid in Fufang Honghua spray.
