Effect of Simvastatin and Coenzyme Q10 on Expression of TGF-β1 in Rats with Chronic Heart Failure

Abstract
OBJECTIVE To study the effect of simvastatin and coenzyme Q10 on expression of transforming growth factor-β1(TGF-β1) in rats with chronic heart failure(CHF). METHODS SD rats were randomly divided into 5 groups: model group, simvastatin group, coenzyme Q10, simvastatin combined coenzyme Q10 group and sham-operated group. The CHF model was established with coarctation of abdominal aorta(CCA). Hemodynamic of left ventricular end-diastolic diameter press(LVEDP), left ventricular pressure rise and fall (±dp/dtmax) was tested; HE and Masson’s trichrome staining was carried out for morphology. Expression of TGF-β1 in left ventricle was assayed by SP immunohistochemistry. RESULTS Compared with model group, LVEDP in other groups was significantly lower(P<0.01), ±dp/dtmax significantly higher(P<0.01); in addition, protein expression of TGF-β1 in left ventricle tissue was also reduced, while simvastatin combined coenzyme Q10 had more obvious effects on improving heart fouction of rats with cardiac remodeling and decreasing expression of TGF-β1 protein. CONCLUSION Simvastatin combined with coenzyme Q10 simvastatin can enhance the efficacy of single treatment on heart fouction, and protein expression of TGF-β1.
