    Determination of Entrapment Efficiency of Oxaliplatin Liposomes by Sephadex Microcolumn combined with HPLC

    • OBJECTIVE To evaluate the quality of oxaliplatin liposomes, and to establish a method to determine the entrapment efficiency of oxaliplatin liposomes. METHODS The microcolumn centrifugation method was employed to separate the free drug from the oxaliplatin liposomes. The content of oxaliplatin was determined by HPLC to calculate the entrapment efficiency. RESULTS A calibrated linear curve of oxaliplatin concentration was within 0.02~1.0mg.mL-1(r=0.9999). The results showed that the average entrapment efficiencies of three batches of oxaliplatin liposomes were 56.6%, 57.5% and 60.0%, respectively. CONCLUSION The method was simple and rapid, and it can be used to determine the entrapment efficiency of oxaliplatin liposomes accurately.
