Determination of Levofloxacin Hydrochloride in Microspheres by HPLC

Abstract
OBJECTIVE To establish an HPLC method for determination of levofloxacin hydrochloride in chitosan microspheres. METHODS The chromatographic analysis was performed on a ZORBAX Eclipse XDB-C18 column (4.6 mm×150 mm, 5 μm) at 30 ℃. The mobile phase was a mixture of 0.04 mol·L-1 phosphoric acid (adjusted to pH 4 with triethylamine) and acetonitrile (85∶15). Flow rate was 1.0 mL·min-1 and UV detection wavelength was 293 nm. RESULTS Good liner relationship was obtained in the range of 1.021-51.15 μg·mL-1(r=0.999 6). Both intra- and inter-day precision RSD were less than 2%(n=5). The average recovery rate was 99.1% with RSD of 0.72%. CONCLUSIONS The established method is sensitive, reliable and suitable for quantitative analysis of levofloxacin hydrochloride in chitosan microspheres.
