Abstract
OBJECTIVE Determination of oleanolic acid and orsolic acid in Pterocephalus hookeri from different batches and different origins. METHODS The separation was performed on an ODS column with the mobile phase of CH3OH-0.1 mol·L-1 ammonium acetate solution. The flow rate was 0.8 mL·min-1. The detection wavelength was 210 nm and the column temperature was 30 ℃. RESULTS The oleanolic acid and orsolic acid linearity range respectively was 1.728-25.92 μg(r=0.999 9), 2.188-32.82 μg(r=0.999 9), the average recovery of oleanolic acid was 99.9%, RSD was 2.12%, the ursolic acid average recovery was 99.9%, RSD was 1.66%. From the results obtained in this study, contents of oleanolic acid and orsolic acid differed from Pterocephalus hookeri collected from different batches and regions. And the contents of oleanolic acid and orsolic acid in Pterocephalus hookeri from Gansu, Qinghai, Xining were higher than others. CONCLUSION This method is stable, reliable, reproduceible. The experimental results supply the basic information for medicinal investigation and the product development of Pterocephalus hookeri.
